If you have 3-4 days, you could probably spend two of those exploring Edinburgh - the castle, the Princes Street area, the royal mile, the new town and so on. Since you can't drive, you could consider taking the train right from Edinburgh city centre to Glasgow or Stirling for a day trip. Stirling is nice and also has an impressive castle and lovely scenery, but it's not a big place. You could easily spend a whole day wandering around Glasgow though - amazing shops, restaurants, pubs, historical buildings (and museums and galleries too!). Stirling is about 25 minutes from Edinburgh by train. Glasgow is 45-50 minutes away.

Good advice from Andrew above. If you would like to see some of the best of Scotland's beautiful natural landscape I would suggest taking the train from Glasgow Queen Street Station to Fort William or Oban, a really gorgeous scenic journey www.scotrail.co.uk

If you only have 3/4 days, spend it in Edinburgh. It's a beautiful city - visit Holyrood, Duddingston, Princes Street, Calton Hill, Arthurs Seat. If you are interested in gardens and museums, it is difficult to advise any further as the city is full of these places and beautiful buildings. If you find you have had enough of sightseeing, take the train to Aberdeen for the day to explore a quite different aspect of Scotland's architecture.
