Can i get on the E train from Penn Station (New York City)?

Yes. You take the E on the uptown side to Lexington Avenue-53rd Street and go through a passageway to get to the number 6 train. It is a fairly long passageway, but just follow the signs and you'll be able to make the transfer.

Yes, after you get off Long Island Railroad, New Jersey Transit, and Amtrak at Penn Station, follow the sign to 8th Av (A)(C)(E) Subway Lines. Then take Queens-bound (E) toward Jamaica Center to Lexington Av-53rd St where (6) train is available at 51st St platform via transfer passegeway. During the weekend, when there is construction work, (E) line might be rerouted to different line for portion of line, etc.
