The quick and short answer is yes. However there is a couple of other points that should be made. One is that answer isn't universally true. While most hotels will accept a prepaid card to hold a room, not all will (this is less of an issue than in the past). The other point is when you use a prepaid or debit card a hotel will have the card company set aside about 150% of the charge amount until paid. This is money you won't have available to be use for other needs. While this isn't really a problem with a credit card, this could be with a prepaid one. It is even more of an issue when you check in, as the hotel will set aside 150% of the total expected bill for your stay right then and there. If you are staying more than one day, this could be a lot of money not at your disposal.
