MLS teams do hold open tryouts but as of right now only the Portland Timbers and Sporting KC have announced them for next year from what i can find here are the sites http://www.portlandtimbers.com/tryouts and http://www.sportingkc.com/2012-open-tryouts. For your next question yes MLS clubs do have youth academy teams some might not but most of them do. Hope this helps.
