I have lost my certificate of Naturalization and haven't made a passport either, but currently trying to apply for a job that requires US citizenship. Is it possible for my employer to find out rather I am a US citizen or not by my social security number?

It's possible IF your employer uses e-verify which is a service of the department of homeland security. HOWEVER, if US citizenship is a requirement of the job, it will be necessary to prove it up with your certificate of naturalization. Report the old one lost or stolen and get a copy of it. It's not that hard.
