Good backpacks for long travel?

What's the difference between hiking backpacks and wheel-free travel bags/luggage?

An typical hiking backpack would be a bigger size backpack with suspension system, hip belt for better comfort during hike. Whereas a wheel-free luggage can be a duffle bag, laptop bag or a sling bag. The only way in which hiking bag differs with wheel-free luggage is toughness, usability in extreme condition.
