I would like create an database-driven online directory website for my Alma Mater which has checkable boxes, filters, and sortable fields where people can search by fields such as graduation year, geographic location, profession, etc. Does anyone know a good place to start?

You should always start with a project plan. Describe the website's functionality. How should it work? What should it look like? If the user clicks on X, it should do Y. Write all those actions down and try to come up with some kind of interaction design. Only when you've figured out what the website should do, can you start thinking about how you're going to program that stuff.
