How much time does it take to get a single DNA test done? How it is done?

Like whether two DNA samples match, whether there is a familial connection etc. Please discuss how the test is done in basic terms. I frequently see test results obtained rather quickly in movies, TV shows. What is realistic duration of the procedure?
