There are two major tracks of applied statistics: experiment design and data mining. For the former, you're going to be asked about how you would design and analyze an experiment, and what sort of sampling schemes you might need to use. For the latter, you're going to be asked about how you would go about building a classifier for some problem. In both cases the difficulty ranges from simple textbook problems to more complicated situations that require some thought.
