The corridors in the Vatican Museum are a total of 4 miles in length. You can't possibly see everything in 4 hours. As others have hinted at, it is important to have your ticket and reserve your time beforehand. You don't want to waste any time standing in line. You may want to do some research first to focus on a few specific areas. To be honest, the Sistine Chapel may be a bit of a disappointment when you see it in person. It is incredibly crowded, and very noisy. It is also very dark. Lights can damage the paintings. You may have trouble seeing any details. The Basilica is free and is open all day. You can probably see most of what you need to in about an hour. Do not miss Michelangelo's Pieta. It is the most beautiful man-made object I have ever seen.

Go at lunch time, the queues are minimal, and the place is practically deserted, though you could spend much longer than 4 hours there, it's pretty wonderful, as long as you ignore the long history of oppression. Enjoy.
