Answer:
It is very hard to answer without knowing your preferences: Germany is very different, also in mentality of the and therefore the cities have a also a different atmosphere: If you want to see big cities and you want to see museums you can visit Berlin, but also Cologne (with its cathedral), Frankfurt or Munich, if you want to see the "romantic" cities you can go to Heidelberg, Rothenburg and in addition cities with an historical heritage I would recommend Luebeck old Hanse town), Muenster, Trier (2000 year old historical building), Freiburg (black forest and famous cathedral), Regensburg (mediaval old town), Bamberg, Würzburg and not to forget Dresden. In addition it depends, how lond you want to stay: The "three day visitors" make the classical "American" trip: Cologne, Rhinevalley, Heidelberg, (maybe Black Forest) Rothenburg, Munich. This short trip covers more the expectation (or better the preconceptions), but it is not the real Germany. A good overview you can find on http://www.germany-tourism.de/ENG/destination_germany/master_tlstadt.htm#top, where you can find a short description of many cities; Click on their names on the left side. But be careful: Not every city mentioned there is really worth to see, because this offical website consider every state (Germany is a federal republic) in the same way.

My Personal Favorites are Berlin and Frankfurt. I LOVE big cities with tons of culture and fashion and both Berlin and Frankfurt provide that. Berlin is more fun in my opinion, but I really like how modern Frankfurt is. Düsseldorf is also a recommended city, it's so pretty in Düsseldorf : )

BERLIN of course!! I think there is no other city where you can see so many different things and styles and collect so many impressions on just one day. Although I have to admit that Berlin is soo special - when you have been there you would still not know what germany is like. At all. Berlin actually feels like an own country! :)
