What are the best ways in which one could use his/her profound resources in the travel sector to start a business/start-up?

If someone has very good resources and goodwill of many influential people in the travel sector and can arrange for finances, what could be the best way(s) in which s/he could start something new - a business or a start-up - aside from the general options of a travel website/travel agency?

That really depends on the type of people you are in touch with. Are they hoteliers? In-bound operators? Airlines? Out-bound agencies? Tourist boards? etc... The travel business is a people business first and foremost, so curating influential contacts is always a good idea. The scope of potential business models you could come up with is vast in such a large, fragmented industry. In-bound operators: A packaged tour e-commerce site (as you mentioned), an aggregator of tours, a marketplace connecting in-bound operators with end consumers (remember end-consumers purchase from the out-bound operators who purchase from the in-bound operators, so why not cut out the middle man) Airlines: an application that gives you information on the aircraft (i.e Seat Guru), a new way of aggregating flights (i.e Hipmunk) Out-bound agencies: Franchising and buying into their purchasing power, representation in local markets, white labeling products and selling online Hoteliers: review sites, room bookings, focusing on a niche booking engine (B&B's, exotic) or maybe something a little different like Air BNB Tourist boards: Representation, travel advice websites/travel blogs Aggregator: Travel organisers that could rival Trip It, guides for cities that you could put on an app and sell to airlines, airport maps and routing calculators This is just a few ideas I came up with quickly, I am sure there are other stakeholders and concepts that are more innovative. I believe Yousef has given a good answer, but I would like to add traveler security. I believe that is an area that will become increasingly important this year and into the near future. The poor world economy has created a lot of social unrest and it looks like Asian countries, including India, Bangladesh and Pakistan, are headed for more problems. There is tension between N. Korea and S. Korea and Japan, and China and Japan are also in a tense situation. Russia is experiencing a rise in social unrest and there is tension with Ukraine. That is only the major ones outside the Middle East. I would look to providing a truly advanced traveler security system.
