What major American businesses (esp. retail and restaurant chains/franchises) use Pay with Square in their nationwide stores?

It'd be interesting if any do at all. Unless they are independently operated. Square's rates are not competitive for high volume businesses. If a big retailer were to sign Square would have to give them some sort of special rate. Which maybe they are totally willing to do. My sense is that they are targeting small business (or micro businesses) that never processed credit cards before. Square is giving them an easy way in. It's a huge win for everyone involved because it's opening a market to credit cards that wasn't there before. That's why you see the big investments by the card companies in Square.
