If I add an annotation on top of a previously uploaded video on Youtube and re-publish the video, will it delete the video view count I already have on that particular video?

Annotations don't cause a YouTube video to be republished, you are simply making an edit to the existing information about the video, just like if you modified the description or tags. The view count is not changed by adding annotations.
