The process of how DNA testing clinics determine paternity takes literally only a few hours; it's just that a lot of these places could be busy at certain times and could take longer to get everything done. The clinic in my link says that they take 2 days. Some paternity clinics even post their results online and thus the turnaround time is even shorter. The actual sample of DNA that they need will usually come from a gentle swab from the inside of the cheek, or a small sample of hair.
